Description
While they were statistically shown to be present in evolutionary data,
large cooperative units, sometimes referred to as "sectors" have not be
observed in Direct Coupling Analysis (DCA) of protein multiple
sequence alignment. We show that due to the sampling limitations of
evolutionary trajectories, these units are bound to be highly conserved
and as such invisible to analysis that is focusing on correlations. We
propose a practical way to quickly augment the DCA procedure that
would allow for discerning the sectors by factoring in the conservation
pattern, similar to the Statistical Coupling Analysis (SCA), which was
the original method of observing cooperative units.
